  2. A combination of galacto-oligosaccharides and Lactobacillus GG increases bifidobacteria to a greater extent than Lactobacillus GG on its own

    The aim of the study was to compare the effects of a combination of Lactobacillus rhamnosus GG (LGG) and galactooligosaccharides (GOS) with LGG on its own on the intestinal microbiota. Forty-four healthy adults were randomised for a double-blind two-period cross-over study. There were two 3-week study periods with a 4-week wash-out period in between. The subjects ingested daily milk-based fruit juice containing either LGG alone or LGG plus GOS. At the end of the study periods the amount of bifidobacteria was significantly greater after the ingestion of LGG plus GOS compared with LGG alone (9.54 vs. 9.30 log(10) cfu/g, p=0.023). No significant differences were seen in the amount of LGG, lactobacilli or pH. Nor did defecation frequency, consistency of stools or ease of defecation differ between the two study periods. However, though neither the occurrence nor the sum of gastrointestinal symptom scores was significantly different between the treatments, the intensity of flatulence was significantly greater after ingestion of both LGG and GOS (17.0 vs. 10.0, p = 0.018). Ingestion of both LGG and 2 g of GOS increases the bifidobacteria more than LGG on its own and thus GOS clearly has a prebiotic effect.

  5. AMPA/kainate receptor-mediated up-regulation of GABA(A) receptor [delta] subunit mRNA expression in cultured rat cerebellar granule cells is dependent on NMDA receptor activation

    We have studied the effects of AMPA/kainate receptor agonists on GABA(A) receptor subunit mRNA expression in vitro in cultured rat cerebellar granule cells (CGCs). Kainate (KA) (100 mu M) and high K+ (25 mM) dramatically up-regulated 8 subunit mRNA expression to 500-700% of that in control cells grown in low K+ (5 mM). KA or high K+ had no effect on the expression of the other major GABAA receptor subunits alpha 1, alpha 6, beta 2, beta 3 or gamma 2. Up-regulation of delta mRNA was also detected with the AMPA receptor-selective agonist CPW-399 and to a lesser extent with the KA receptor-selective agonist ATPA. AMPA/kainate receptor-selective antagonist DNQX completely inhibited KA-, CPW-399- and ATPA-induced 6 mRNA upregulation indicating that the effects were mediated via AMPA and KA receptor activation. NMDA receptor-selective antagonist MK-801 inhibited 76% of the KA- and 57% of the CPW399-induced delta up-regulation suggesting that KA and CPW-399 treatments may induce glutamate release resulting in NMDA receptor activation, and subsequently to 6 mRNA upregulation. In CGCs, delta subunit is a component of extrasynaptic alpha 6 beta delta receptors that mediate tonic inhibition. Up-regulation of 6 during prolonged glutamate receptor activation or cell membrane depolarization may be a mechanism to increase tonic inhibition to counteract excessive excitation.   8. Acute effects of AMPA-type glutamate receptor antagonists on intermale social behavior in two mouse lines bidirectionally selected for offensive aggression

    Involvement of AMPA-type glutamate receptors in the regulation of social behavior has been suggested by experiments with mice deficient for the GluR-A subunit-containing AMPA receptors showing reduced intermale aggression. In the present study, effects of AMPA receptor antagonists on mouse social behavior towards unfamiliar Swiss-Webster males on a neutral territory were tested using male subjects from the Turku Aggressive (TA) and Turku Non-Aggressive (TNA) mouse lines bidirectionally selected for high and low levels of offensive aggression. The drugs were the competitive antagonists 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX) and 2,3-dioxo-6-nitro-1,2,3,4-tetrahydrobenzo[f] quinoxaline-7-sulfonamide (NBQX), and the non-competitive antagonist 4-(8-methyl-9H-1,3-dioxolo[4,5-h][2,3]benzodiazepin-5-yl)-benzenamine (GYKI 52466). In TA mice, CNQX and NBQX decreased the biting component of aggressive structure, while GYKI 52466 suppressed all aggressive manifestations. All drugs increased anxiety-like behavior towards the partner. In TNA mice, NBQX activated mouse social behavior and ambivalent aggression, while CNQX and GYKI 52466 only increased anxiety. Thus, AMPA receptor antagonists affect aggressive behaviors in TA mice supporting the idea that AMPA receptors are involved in the modulation of agonistic impulsive behavioral pattern. GYKI 52466 appeared to be the most selective and efficacious in suppressing the aggression. (c) 2007 Elsevier Inc. All rights reserved.
